Application

The Anti-His6(2) antibody provides specific and sensitive detection of histidine-tagged proteins, regardless of the vector used for expression. The antibody is suitable for immunoblotting, immunoprecipitation, immunoassays (ELISAs), immunocytochemistry, immunofluorescence, and flow cytometry. When used in combination with metal chelate affinity chromatography, efficient purification is coupled with sensitive detection of a tagged, recombinant protein.

Product Description

Specificity:
The Anti-His6 (2) antibody recognizes an epitope of six consecutive histidine residues (His6) in natural and recombinant proteins. The antibody reacts with native and denatured histidine-tagged fusion proteins independent of the location of the histidine tag in the protein sequence; however, it preferentially recognizes the N-terminal His6 epitope with high sensitivity.

An N-terminally His-tagged GFP-fusion protein (His6-GFP) was serially diluted in an untransformed E. coli crude extract (2 μg total protein per lane) and indirectly detected with Anti-Mouse-POD (50 mU/ml). The Anti-His6 (2) antibody was diluted 1:3000 for colorimetric detection (a) using BM Blue POD Substrate* and
1:10,000 for chemiluminescent detection (b) using Lumi-LightPLUS Western Blotting Substrate*.
