Application

The incorporation of 7-deaza-2'-deoxyguanosine-5'-triphosphate (7-deaza-dGTP) into DNA can eliminate spurious GC-hydrogen bonding and relax secondary structures, an effect that may be useful in all types of dideoxy sequencing and polymerization techniques. For example:

  • Sequencing: Use 7-deaza-dGTP) instead of dGTP in primer extension reactions (e.g., Sanger sequencing) to better resolve GC-rich regions on gels.
  • PCR: Partial substitution of 7-deaza-dGTP for dGTP in PCRs can improve the product yield when the template is GC-rich (i.e., has extensive secondary structures).

Notes

When PCR products contain 7-deaza dGTP, they do not stain well in ethidium bromide, presumably because stacking of adjacent bases in the products is diminished. 

PCR amplification efficiency is generally not affected by 7-deaza-dGTP.
