Application

Characterization and analysis of carbohydrate structures by the specific binding of selected lectins. In addition to the differentiation of glycoproteins bound to nitrocellulose membranes, the lectins are also suited for the identification of carbohydrate structures on tissue sections.

Principle

Detection of digoxigenin-labeled lectins bound to specific carbohydrate structures on blots. Detection is performed using anti-digoxigenin alkaline-phosphatase conjugates. The digoxigenin-labeled lectins enable differentiation between complex and high-mannose chains, differentiation between a(2-3) and α(2-6) linkage of terminal sialic acids, and identification of the core disaccharide Galβ(1-3)GalNAc of O-glycans.

Quality

Function tests: With the lectins GNA, SNA, MAA, and DSA 0.1 µg, and with the lectin PNA 1 µg of the control glycoproteins are detectable after a 1-min incubation in the staining solution.
